摘  要:目的:探讨PTEN基因的表达在人大肠癌发生发展及浸润转移过程中的作用。方法:应用RT-PCR和Western blot的方法,对75例人大肠癌及癌旁组织中PTEN mRNA和蛋白的表达进行分析。结果:PTEN mRNA在大肠癌癌旁肠粘膜,早期癌及进展期癌中的阳性表达率分别为93.3%,75.0%和47.6%,PTEN蛋白在上述组织中的表达率则分别为96.0%,66.7%和49.2%。PTENmRNA和蛋白的表达均与大肠癌癌细胞的分化程度,肿瘤的Dukes分期及有无淋巴结转移密切相关(P<0.05)。PTEN mRNA在进展期癌中的表达明显低于早期癌(P<0.05)。PTENmRNA与PTEN蛋白的表达呈显著正相关(r=0.99,P<0.01)。结论:PTEN mRNA与蛋白表达的减少与大肠癌的发生和进展密切相关。Objective: To investigate the relationship between PTEN expression and the carcinogenesis of colorectal mucosa, invasion and metastasis of colorectal carcinoma. Method: The mRNA and protein expressions of PTEN were evaluated by reverse transcription polymerase chain reaction ( RT - PCR) and Western blot in 75 cases of colorectal carcinomas and the adjacent normal mucosa. Result: The positive rates of PTEN mRNA expression were 93.3%, 75.0 and 47.6% in adjacent normal mucosa, early and advanced colorectal carcinoma, respectively. The positive rates of PTEN protein expression were 96.0%, 66.7% and 49.2% in corresponding tissues. PTEN mRNA and protein expression was significantly correlated with cell differentiation ( P 〈 0.05 ), tumor Dukes stage and lymph node metastasis of colorectal carcinomas ( P 〈 0. 05 ). Significant difference in PTEN mRNA expression was found between early colorectal carcinoma and advanced colorectal carcinoma (P 〈0.05). The expression of PTEN mRNA was positively associated with that of PTEN protein ( r = 0. 99, P 〈 0. 01 ). Conclusion: Decreased expressions of PTEN mRNA and protein were significantly correlated with carcinogenesis and progression of the colorectal carcinoma.
