摘  要:从土壤中筛选分离出的产纤维素酶菌株S26(经中国科学院微生物所鉴定为产黄纤维单胞菌Cellu-lomonas flavigena),测定酶活为25.86U/mL,以此为出发菌株,经UV反复诱变处理,多代选育,筛选出1株纤维素酶高产突变株UY-4,酶活力达87.92U/mL,是出发菌株的3.4倍,而且遗传性能稳定。对UY-4产酶影响因素的研究表明,产酶最适条件为稻草与麦麸3:2、接种量10%(体积与质量比),(NH4)2SO40.5%、起始pH7.4、35℃、培养72~84h。A cellulase-producing strain S26 was screened from soil (identified as Cellulomonas flavigena by Institute of Microbiology, Chinese Academy of Sciences ) with enzymatic activity tested at 25.86 U/mL. And this strain was taken as starting strain and repeatedly treated with UV to mutate and bred for several generations, a cellulase-high- producing strain UY-4 was obtained with enzymatic activity at 87.92 U/mL, that was 3.4 times that of the starting strain, furthermore, it was genetically stable. Study on the influence factors of enzyme production of UY-4 indicated that the most suitable conditions for the cellulase production were straw : wheat bran = 3 : 2, 10% of inoculation (volume/weight) , 0.5 % of ( NH4 ) 2 SO4, 35 ℃ , initial pH at 7.4, culture for 72 - 84 hours.
