摘  要:目的建立三棱超临界CO2萃取物中芦丁的鉴别和含量测定方法。方法在聚酰胺膜上点样,以80%乙醇作为展开剂鉴别芦丁,采用SymmetryC18色谱柱,以乙腈-四氢呋喃-0.2%枸橼酸(14:1:85)为流动相,柱温25℃,在流速为1.0ml·min^-1,360nm的检测波长下,测定芦丁含量。结果在三棱的超临界萃取物中含有芦丁成分,建立的液相色谱方法在芦丁浓度为0.00340~0.02432mg·ml^-1的范围内线性关系良好,样品在8h内稳定,平均加样回收率为98.56%,以峰面积作精密度实验,RSD为1.38%。结论三棱的超临界萃取物中含有芦丁,建立的高效液相色谱方法线性关系良好.精密度和回收率高,适合于三棱中芦丁含量的测定。Objective To explore whether there is rutin in SFE extract of Sparganium stoloniferum by TLC and establish a method to determine its content by RP - HPLC. Methods The determination was carried out over a Symmetry C18 column at 25℃. Methods tetrahydrofuran, 0.2% citric acid (14: 1: 85) was used as mobile phase at a velocity of 1 ml · min^-1 and the detection wavelength was set at 360nm. Results Rutin was found in SFE extract of Sparganium Stoloniferum. The calibration curve was linear in the range of 0.003 40 - 0. 02432 mg · ml^-1 for rutin, The sample was stable within 8 hours and the recovery rate of rutin was 98.56 %. And in precision test, RSD = 1.38%. Conclusion There is rutin in SFE extract of Sparganium stoloniferum. The established RP - HPLC method has perfect linear relationship, high precision and recovery rate and it is suitable to determine the content of rutin in SFE extract of Sparganium Stoloniferum.
