摘 要:目的:检测卵巢癌细胞株SKOV-3中人白细胞抗原G(human leucocyte antigen-G,HLA-G)的表达。方法:用逆转录酶聚合酶链式反应(reverse transcriptase-polymerase chain reaction,RT-PCR)、免疫细胞化学法及流式细胞术检测卵巢癌细胞株SKOV-3中HLA-G的表达,以绒毛膜癌细胞株JEG-3中HLA-G的表达为阳性对照。结果:(1)免疫细胞化学显色显示SKOV-3细胞膜及细胞浆内均有HLA-G的表达;(2)RT-PCR结果表明,SKOV-3细胞中有HLA-G mRNA表达;(3)流式细胞术结果提示,SKOV-3细胞中HLA-G阳性率是33.77%。结论:卵巢癌细胞株SKOV-3中HLA-G mRNA及HLA-G蛋白表达为阳性。Objective :To investigate the expression of human leucocyte antigen-G(HLA-G) in ovarian cancer cell line SKOV-3. Methods:The expression of HLA-G in choriocarcinoma cell lines JEG-3 as the positive control, RT-PCR, immunocytochemical method and flow cytometry were used to detect the expression of HLA-G in ovarian cancer cell line SKOV-3. Results: ( 1 ) HLA-G were found at the membrane and the cytoplasm by immunocytochemical method. (2) HLA-G mRNA were found in ovarian cancer cell line SKOV-3 by RT-PCR. (3)The results of flow cytometry showed the rate of the HLA-G-positive tumor cells was 33.77%. Conclusion:The expression of HLA-G mRNA and HLA-G protein in ovarian cancer cell line SKOV-3 are positive.
