摘  要:目的探讨食管鳞癌细胞(ECCs)的干细胞特性。方法将ECCs于无血清培养液(SFM)中培养,将其中可悬浮生长并形成的细胞球取出,制成单细胞悬液,以p75NTR为抗原标记细胞后进行流式细胞仪检测,观察其中p75NTR阳性细胞的比例。结果ECCs可在SFM中悬浮生长并形成细胞球,其形成细胞球的比例为3·6%~17·0%。悬浮生长的细胞中p75NTR阳性细胞比例为16·25%。结论ECCs在SFM中可悬浮生长并形成细胞球,其中的p75NTR阳性细胞具有干细胞的某些特性,其可作为食管鳞癌干细胞的标记之一。Objective To study the properties of p75^NTR positive cells in human esophageal carcinoma cells suspension cultured in a serum-free medium. Methods ECCs were suspension cultured in a serum-free medium and cell spheres were formed. FCM was used to detect theproportion of p75^NTR positive cells of spheres-cells cultured in SFM. Results In serum-free medium ECCs could propagate and form cell spheres successfully. The proportion of cell spheres formed was 3.6 - 17.0%. Using flow cytometry, higher proportions of p75^NTR positive cells were detected in spheres-cells cultured in SFM (16.25%:1.26%, P 〈 0.01 ). Conclusion The p75^NTR positive cells had some properties of stem cells, such as the ability to self-renew and proliferate; p75^NTR may be the candidate of surface markers in cancer stem cells in ECCs.
