摘 要:淋巴管系统是恶性肿瘤转移的重要途径,但是人们对淋巴管生成在恶性肿瘤中的作用机制知之甚少。多种细胞因子参与了调节淋巴管生成的细胞信号传导通路,这些细胞因子包括:VEGF-C,VEGF-D,VEGFR-3,VEGF-A,FGF-2,Prox-1,Ang2和NP-2等;近年来发展起来的多种淋巴管内皮标志,如podoplanin、LYVE-1、Prox-1、VEGFR-3和LVEF等,为研究淋巴管生成在肿瘤中的作用提供了有力的工具;研究发现胃肠道肿瘤的淋巴管主要分布在肿瘤的周边区和正常区,肿瘤癌周组织的淋巴管密度明显高于正常组织的淋巴管密度;最近的研究还表明胃肠道恶性肿瘤的淋巴管生成与肿瘤的复发、转移和总生存率有一定的相关性。Lymph drainage system is one of the most important ways by which tumor metastases. However, the mechanisms beneath the roles of lymphangiogenesis in the metastasis of cancer are not well addressed. The signal pathways regulating lymphangiogenesis involves numerous factors, including VEGF-C, VEGF-D, VEGFR-3, VEGF-A, FGF-2, Prox-1, Ang2, NP-2, et al. Recently developed lymphatic endothelium markers, such as podoplanin, LYVE-1, Prox-1, VEGFR-3, and LVEF, provides powerful tools to explore the roles of lymphangiogenesis in tumor development and progression. Previous studies indicated that lymphatic vessels were mainly seen in peritumoral tissues and normal tissues in gastrointestinal cancers, and lymphatic vessel density in peritumoral tissues was higher than it in normal tissues. Moreover, recent studies showed that the lymphangiogenesis was associated with recurrence, metastasis and overall survival in gastrointestinal cancers.
