摘  要:目的:探讨宫颈鳞癌组织中高迁移率族蛋白HMGB1表达情况及其与肿瘤分化程度、大小、浸润和转移的关系。方法:应用组织芯片免疫组织化学方法检测30例宫颈原位癌、90例无转移的宫颈鳞癌、30例有转移的宫颈鳞癌和30例正常宫颈鳞状上皮中HMGB1表达情况,并应用Westernblot技术定量检测其表达情况。结果:HMGB1在宫颈鳞癌组织中阳性表达为58.67%(88/150),明显高于正常宫颈鳞状上皮的16.66%(5/30),差异有统计学意义,P<0.01。HMGB1阳性率与肿瘤分化程度无关,P>0.05;而与肿瘤的大小、浸润及转移呈正相关,P<0.01。结论:HMGB1在宫颈鳞癌组织中尤其是发生转移的宫颈鳞癌组织中呈强阳性表达,可作为宫颈鳞癌浸润、转移及预后的重要判定指标之一。OBJECTIVE: To investigate the expression of HMGB1 (high mobility group boxl, HMGB1 ) in human CSEC (cervical squamous epithelial carcinoma) and to explore its relation to differentiation degree, size, invasion and metastasis of CSEC. METHODS: The immunohistochemical staining of tissue microarrays was applied (30 cases of carcinoma in situ, 90 cases of invasive CSEC with non metastasis, 30 cases of invasive CSEC with metastasis and 30 cases of normal cervical squamous epithelial tissues, respectively) as well as Western blot analysis to detect the expression of HMGB1 in CSEC. RESULTS: The positive expression rate of HMGB1 was 58.67%(88/150)in CSEC, showing a significant difference between it and the normal cervical squamous epithelial tissues (16.66%, 5/30). The expression of HMGB1 was correlated with tumor size,invasion and metastasis of CSEC(P〈0. 01 ), but had no relation with the differentiation degree (P〉0.05). CONCLUSION : The over expression in CSEC might be one of the parameters for determine tumor invasion, metastasis, and prognosis.
