卵巢原发癌和转移癌组织中VEGF上调对Bcl-2和Survivin表达影响的相关性研究

摘　　要：目的:分析卵巢原发癌和转移癌组织中血管内皮生长因子(VEGF)的过表达,对凋亡抑制因子Bcl-2、Survivin表达的影响以及临床病理特征相关性。方法:应用免疫组化SP法检测VEGF、Bcl-2蛋白,原位杂交技术检测Survivin mRNA在正常卵巢组织、良恶性卵巢肿瘤中的表达。结果:VEGF、Bcl-2和SurvivinmR-NA在卵巢正常组织、良性肿瘤及卵巢恶性肿瘤(原发癌和转移癌)中的表达,差异均有统计学意义,P<0.01。卵巢恶性肿瘤中3种基因表达与淋巴转移有关,P<0.05,卵巢原发癌组织中VEGF的表达与临床分期有关,P<0.05。Bcl-2、Survivin与VEGF的表达呈正相关,r值分别为0.329和0.317,P值均<0.05。结论:VEGF、Bcl-2和Survivin在卵巢原发癌和转移癌中均为过表达,与淋巴转移关系密切。VEGF的表达上调,可能促进Bcl-2、Survivin的过表达,并在卵巢恶性肿瘤发生发展和转移中发挥重要的协同效应。OBJECTIVE： To discuss the correlation between the over expression of vascular endothelial cell growth factor （VEGF） and the expression of Bcl-2, survivin in primary and metastatic ovarian carcinoma and the clinicopathologic characteristics. METHODS： The expressions of VEGF and Bcl-2 were detected by the immunohis- tochemical assay. The expression of suvivin was detected by in situ hybridization in normal ovary, ovarian benign and malignant tumor and metastatic ovarian carcinoma. RESULTS： The positive expression of VEGF,Bcl-2 and Suvivin in ovarian carcinoma （primary and metastatic） were statistically different from those in normal and benign tumor of ovary,P〈0. 01. The expressions of all the three genes were associated with lymph node metastasis,P〈0. 05. The positive expression of VEGF was associated with clinical stage in primary ovarian carcinoma, P〈0. 05. The analysis showed that Bcl-2 and Survivin were positively associated with VEGF, with which r were 0. 329 and 0. 317 respectively, and P were both 〈0.05. CONCLUSIONS：VEGF,Bcl-2 and suvivin are all over expression in primary and metastatic ovarian carcinoma, and have close correlation with lymph node metastasis. The up-regulation expression of VEGF may promote the over expressions of Bci-2 and survivin, which plays an important cooperative role in the onset and progression of ovarian malignant tumor.
