摘  要:[目的]探讨nm 23-H1基因蛋白及核增殖抗原(PCNA)在喉鳞状上皮癌中的表达及其与临床病理特征的关系.[方法]选择喉鳞状上皮癌标本50例,另选择喉癌旁黏膜标本10例,采用免疫组织化学SP法进行染色,观察nm 23-H1基因蛋白和PCNA的表达情况,分析PCNA和nm 23-H1基因蛋白的表达与喉鳞状上皮癌的病理类型、TNM分级、临床分期及淋巴结转移情况的关系.[结果]nm 23-H1基因蛋白在喉鳞状上皮癌中的表达率为68%,在喉癌旁组织中为90%,两者间比较有显著性差异;nm 23-H1基因蛋白的表达强度与喉鳞状上皮癌的TNM分级、临床分期和淋巴结转移情况呈显著相关.PCNA在喉鳞状上皮癌中的表达率为74%,在喉癌旁组织中为40%,两者间比较有显著性差异;PCNA的表达强度与喉鳞状上皮癌的组织学类型、TNM分级、临床分期和淋巴结转移情况呈显著相关.[结论]nm 23-H1基因蛋白和PCNA的表达强度可作为判断喉鳞状上皮癌生物学特性的主要指标,nm 23-H1基因蛋白低表达和PCNA高表达与喉鳞状上皮癌生物学特性有关.喉鳞状上皮癌组织中nm 23-H1基因蛋白和PCNA的表达呈负相关.OBJECTIVE To study the expression of nm23-H1 and proliferating cell nuclear antigen(PCNA) in laryngeal squemous cell cacinoma(LSCC) and its relationship with the characteristis of clinicopathology.METHODSThe expression of nm23 and PCNA was detected by immunohistochemical SP method in 50 cases of LSCC and 10 cases of normal laryngeal mucosa,and the relationship among expression of PCNA and nm23-H1 gene protein and pathologic type,TNM grade,clinical stage,and metastasis of lymph node.RESULTSThe positive rate of nm23 H1 was 68% and 90% in LSCC and normal larynx mucosa,respectively,and there was a significant difference between two groups.The intensity of positive expression of nm23 H1 was significantly related with the TNM grade,clinical stage and metastasis of lympho node in LSCC.The positive rate of PCNA was 74% and 40% in LSCC and normal larynx mucosa,respectively,and The intensity of positive expression of PCNA was significantly related with the histopathological type,TNM grade,clinical stage and metastasis of lympho node in LSC.CONCLUSIONThe intensity of positive expression of nm23 H1and PCNA can be as the important index for judging the biological characteristics of LSCC.The low expression of nm23 H1 and high expression of PCNA is related with biological characteristics of LSCC.There is negative correlation between the expression of nm23 H1 and PCNA protein in LSCC.
