摘 要:目的:检测皮肤鳞状细胞癌(SCC)和基底细胞癌(BCC)组织中环氧合酶-2(COX-2)和缺氧诱导因子-1α(HIF-1α)的表达,分析皮肤癌组织中2者的相关性及与血管生成的关系。方法:采用免疫组织化学方法检测25例SCC、25例BCC及23例正常皮肤组织中COX-2、HIF-1α和CD34的表达,并计数CD34标记的微血管密度(MVD)。结果:SCC和BCC组织中COX-2和HIF-1α蛋白阳性表达率及CD34-MVD均高于正常皮肤组织(P<0.05)。COX-2和HIF-1α的表达相关(P<0.05)。SCC和BCC组织中COX-2、HIF-1α表达阳性者的MVD值均高于表达阴性者(P<0.05)。结论:SCC和BCC组织中COX-2与HIF-1α可能协同促进肿瘤新生血管生成,从而促进SCC和BCC的生长、浸润或转移。Aim : To investigate the expression of cyclooxygenase-2 ( COX-2), hypoxia-inducible factor-1α (HIF-1α) and the correlation with angiogenesis in cutaneous squamous cell carcinoma (SCC) and basal cell carcinoma (BCC) tissue. Methods:The SP immunohistochemistry method was used to detect the expression of COX-2, HIF-1α and CD34-MVD in 25 cases of SCC, 25 cases of BCC, and 23 cases of normal skin tissue. Results: The positive expression rates of COX- 2 and HIF-1α,and MVD marked by CD34 in SCC and BCC tissue were significantly higher than those in normal skin tissue (P 〈0.05). The expression of HIF-1α was related to the expression of COX-2 in SCC and BCC tissue(P 〈0.05). The cases with positive expression of COX-2 and HIF-1α had higher MVD (P 〈 0.05). Conclusion :There are overexpression of COX-2 and HIF-1α in BCC and SCC. They probably have a synergistic effect in the neovascularization of the tumor, thus promote the carcinogenesis and development of SCC and BCC.
