摘 要:目的:检测全反式维甲酸(ATRA)诱导后HL-60细胞核干细胞因子(NS)mRNA表达的变化,探讨NS mRNA表达与细胞分化之间的关系。方法:从NS基因的3个变异体中筛选出共同序列设计特异性引物,用终浓度为0.1μmol/L、1μmol/L、10μmol/L的ATRA诱导分化HL-60白血病细胞48h,以不加ATRA的HL-60细胞作为对照,应用逆转录-聚合酶链式反应(RT-PCR)检测HL-60细胞NS mRNA表达水平的变化。结果:ATRA诱导HL-60细胞分化后NS mRNA表达均有所降低,0.1μmol/L、1μmol/L、10μmol/L浓度下NS mRNA表达量分别下降3.72%±0.84%、58.70%±4.98%和61.60%±2.26%。其中1μmol/L和10μmol/L组与对照组和0.1μmol/L组比较,差异均有统计学意义(P均<0.05)。结论:NS可能与白血病细胞分化有关,参与调控细胞分化过程。Aim: To determine the expression of nucleostemin (NS) mRNA in HL-60 cells after induced differentiation, and to approach the potential mechanism between NS mRNA and pathogenesis of acute leukemia. Methods:The common sequence was screened out from three variants of NS gene and the primers were designed. HL-60 cells were induced and differentiated by ATRA in 0. 1 μmol/L, 1 μmol/L,and 10 μmol/L concentration groups for 48 hours, and the NS mRNA expression was detected in HL-60 cells by RT-PCR. The same detection was performed in the undifferentiated HL-60 cells of control group. Results : The expression of NS mRNA in different concentration of ATRA decreased than that of control group. After induced and differentiated by ATRA of 0.1μmol/L, 1 μmol/L, and 10 μmol/L concentration, the expression of NS mRNA in HL-60 cells decreased 3.72% ± 0. 84% , 58.70% ± 4.98% , and 61.60% ± 2.26% ,respectively. And NS mRNA expression decreased significantly in 1 μmol/L, 10 μmol/L groups compared with control group and 0.1 μmol/L group ( P 〈 0.05 ). Conclusion : NS may be in relation to the differentiation process of leukemia cells.
