摘  要:应用AFLP分子标记技术对我国三疣梭子蟹黄海和东海两群体共40个个体利用10对选择性引物组合进行了遗传多样性分析。共扩增出411个位点,多态位点333个。黄海和东海群体的多态位点比例,Nei基因多样性指数和Shannon遗传多样性信息指数分别为74.5%、63.1%,0.2553、0.2355,0.3827、0.3497,两群体的遗传多样性均处于同一水平。群体间遗传分化系数Gst、Shannon遗传多样性信息指数表明,两群体的遗传变异主要来源于群体内个体间。UPGMA聚类图及两群体的Gst和遗传距离显示两群体间出现了一定的遗传分化。As an economically important marine species, Portunus triuberbuculatus has been studied intensively on fetation, production, and genetic mark by RAPD; however, little is known on the population quality, genetic diversity and structure. Therefore, to understand the variation in population genetics, 20 individuals were collected in each Qingdao and Zhoushan in 2006 representing the populations of the Yellow Sea and the East China Sea, respectively. Amplified Fragment Length Polymorphism (AFLP) technology was used to analyze the genetic diversity and differentiation of the two natural populations. 411 loci were generated from the 40 individuals using 10 primer combinations, of which 333 were polymorphic (the mean percentage of polymorphism was 81.02%). The proportion of polymorphic loci, the Nei's gene diversity and Shannon's Information Index of the Yellow Sea and East China Sea were 74.5% and 63.1%, 0.2553 and 0.2355, and 0.3827 and 0.3497, respectively. The results show that both populations were at the same high level of genetic diversity. Gst value and Shannon's Information Index indicated that the genetic variation mainly came from different individuals within the population. Gst value, inter-population genetic distance, and UPGMA showed no significant genetic differentiation between the two populations. The distribution of dominant gene frequency revealed, the two populations had similar genetic structure. Therefore, both wild populations had high intra-population genetic diversities and low inter-population differentiation. Protection of their living environment and reasonable use of the resource need to be reinforced.
