摘 要:目的探讨新型的中枢促醒药物莫达非尼对不同程度快速动眼(REM)睡眠剥夺及睡眠恢复后大鼠认知功能的影响。方法成年雄性SD大鼠随机分为对照组、溶媒剂组和莫达非尼组,按REM睡眠剥夺及剥夺后恢复睡眠的时间每组分5个时间点(REM睡眠剥夺1、3、5 d及REM睡眠剥夺5 d后恢复睡眠6、12 h),每个时间点6只大鼠,莫达非尼组予莫达非尼混悬液300 mg·kg^(-1)·d^(-1)灌胃,溶媒剂组予双蒸馏水配置的0.3%羧甲基纤维素钠混悬液16 mL·kg^(-1)·d^(-1)灌胃,对照组不予干预。用改良多平台水环境REM睡眠剥夺法进行REM睡眠剥夺,利用Y迷宫测定大鼠学习、记忆能力。结果对照组REM睡眠剥夺3、5 d时的错误反应次数(EN)明显多于REM睡眠剥夺1 d时(P值均<0.05),莫达非尼组各时间点EN的差异无统计学意义(P值均>0.05)。对照组与溶媒剂组大鼠各时间点EN和总反应时间(TRT)的差异均无统计学意义(P值均>0.05),莫达非尼组大鼠的EN在REM睡眠剥夺后明显少于对照组和溶媒剂组(P值均<0.05),睡眠恢复后差异均无统计学意义(P值均>0.05),莫达非尼组大鼠各时间点,TRT与对照组和溶媒剂组的差异均无统计学意义(P值均>0.05)。结论不同程度的REM睡眠剥夺均会对认知功能造成不利影响,莫达非尼对REM睡眠剥夺后的认知行为下降有改善作用。Objeetive To investigate the effects of modafinil, a novel wake-promoting agent, on the cog nitive function of rats after rapid eye movement (REM) sleep deprivation for different periods and after sleep re covery. Methods Adult SD rats were randomized equally into modafinil group, control group, and carboxy- methylcellulose sodium (solvent) group. Each group was further divided randomly into 5 subgroups, in which the rats were subjected to REM sleep deprivation for 1 day (SDld), 3 days (SD3d), 5 days (SD5d), 5 days with 6 hours recovery sleep (SD5d/RS6h), and 5 days with 12 hours recovery sleep (SD5d/RS12h), respectively, with 6 rats in each subgroups. In the modafinil group, modafinil suspension was intragastrieally administered at 300 mg · kg^-1· d^-1 , the rats in the solvent group received only the solvent diluted with double distilled water at 3 mL/d in the same manner, and no interventions were administered in the control group. The modified multiple platform method (MMPM) was used to establish the REM sleep deprivation model and the cognitive function of the rats was evaluated with the Y maze test. Results In the control group, the rats with SD3d and SD5d showed significantly greater error number (P〈0.05) than those with SDld. No significant difference was found in the error number between the subgroups with modafinil treatment (P〉0.05), nor in the error number or total reaction time between the control group and solvent group at the corresponding time points (P〉0.05). In the modafinil group, the rats with REM sleep deprivation showed significantly fewer errors than those in the other two groups (P〈0.05), but after the recovery sleep, the differences were no longer significant. There were no statistically significant differences in the total reaction time between the 3 groups. Conclusion Sleep deprivation,brief or prolonged, all have adverse effects on the cognitive function, and modafinil may improve the deteriorated cognitive function after sleep
