特定条件下化疗诱导结肠癌细胞凋亡的研究

摘　　要：目的:研究结肠癌化疗药物氟尿嘧啶(5-FU)和奥沙利铂(OXA)分别诱导结肠癌细胞株HT-29细胞凋亡的作用。方法:用80μg/ml5-FU和25μg/mlOXA分别作用HT-29细胞8h,并应用Annexin V-FITC/PI双标记流式细胞术对细胞凋亡进行检测,观察凋亡细胞的比例。结果:80μg/ml5-FU处理8h组与空白对照组比较,凋亡细胞比例上升约9.2%(P<0.01);25μg/mlOXA处理8h组与空白对照组比较,凋亡细胞比例上升约21.1%(P<0.01)。25μg/mlOXA处理8h组与80μg/ml5-FU处理8h组比较,凋亡细胞比例约高于11.9%(P<0.01)。结论:在特定条件下,5-FU和OXA均能明显诱导结肠癌细胞株HT-29细胞凋亡;OXA对结肠癌细胞的诱导凋亡作用要强于5-FU。Objective：To observe the apoptosis-inducing effect of 5-FU and oxaliplatin under specific condition in colon cancer cell line HT-29. Methods：HT-29 cells were separately treated with 80μg/ml 5-FU and 25μg/ml oxaliplatin for 8 hours. And the cellular apoptosis was analyzed by AnnexinV-FITC/PI double staining flow cytometry. Results： HT-29 cell line was detected by AnnexinV-FITC/PI double staining flow cytometry. Compared with the blank group, the proportion of apoptosis cells of the group treated with 80μg/ml 5-FU for 8 hours increased approximately 9. 2% （P 〈0. 01 ）. and the proportion of apoptosis cells of the group treated with 25μg/trd oxaliplatin for 8 hours increased approximately 21.1% （P 〈0.01 ）. Compared with the group treated with 80μg,/ml 5-FU for 8 hours, the proportion of apoptosis cells of the group treated with 25 μg,/ml oxaliplatin for 8 hours increased approximately 11.9% （ P 〈 0. 01 ）. Conclusion ：5-FU and oxaliplatin under specific condition both can induce apoptosis of HT-29 cell line. Oxaliplatin resuhes in stronger effect of apoptosis inducing than 5-FU in colon cancer cells.
