摘 要:目的:检测PTTG在甲状腺乳头状癌组织中的表达,探讨其与甲状腺乳头状癌的关系。方法:在52例甲状腺乳头状癌组织及其癌旁组织中分别应用原位细胞凋亡检测(TuNEL)试剂盒测定原位细胞凋亡指数(apoptosis index,AI),应用免疫组化方法检测PTTG,应用原位杂交方法检测PTTG mRNA表达。结果:PTTG及PTTG mRNA在甲状腺乳头状癌组织中表达比相应癌旁组织显著升高(P=0.0091,P=0.0012):PTTG及PTTG mRNA灰度值与AI正相关(r=0.447,P=0.003;r=0.564,P=0.000)。结论:PTTG及PTTG mRNA表达升高可能参与甲状腺乳头状癌的发生发展,检测PTTG及PTTG mRNA对于甲状腺乳头状癌与良性组织鉴别诊断具有临床参考意义。Objective:To study the relationship between PTTG and papillary thyroid carcinoma by combining assays of PTTG and PTTG mRNA in papillary thyroid tissue. Methods: Apoptosis index was evaluated by TUMEL method, the expression of PTTG and PTTG mRNA were detected by immunohistochemistry and in situ hybridization in 52 papillary thyroid carcinoma cases and tissue around carcinoma. Results: The higher level of PTTG and PTTG mRNA in papillary thyroid carcinoma was found compared with tissue around carcinoma( P = 0. 0091, P = 0.0012 ) ; There had positive relationship between the expression of PTTG, PTTG mRNA and AI ( r = 0. 447, P = 0. 003 ; r = 0. 564, P = 0. 000 ). Conclusion: PTTG may participate in proceeding of papillary thyroid carcinoma, it is sug- gested that combined detections of PTTG and PTTG mRNA may have important reference significance on the differentiation of benign and malignant papillary thyroid diseases.
