摘　　要：目的探讨新生SD大鼠视网膜Mller细胞原代培养的方法。方法用酶消化法分离制备视网膜细胞悬液,采用分次贴壁和反复吹打法分离并纯化Mller细胞,进行形态学观察及免疫组织化学染色。结果光镜下培养细胞体较大,呈扁平不规则形状;免疫组织化学染色显示95%以上的原代细胞神经胶质纤维酸性蛋白染色阳性;电镜下细胞内可见丰富的线粒体、高尔基体和大量直径8～10nm的微丝结构。结论分次贴壁和反复吹打法是一种简单可靠的视网膜Mller细胞原代培养的方法。Objective To investigate a met-hod for primary culture of newborn SD rats＇ retinal Müller cells. Methods The cells suspension of retina was obtained by using enzyme digestion method,retinal Müller cells were cultured and purified by the multiple attaching and shaking method, and identified by the morphological observation and immunohistological staining. Resuits More than 95% primary cells showed positive reaction to glial fibrillary acidic protein with immunohistoehemical staining. The morphology showed it contrained many intermedicate filaments with 8 - 10 nm in diameter with electron microscope. Conclusion The multiple attaching and shaking method is a rapid and reliable way for culturing Müller cell.
