摘  要:目的本实验探讨了玻璃体内注射地塞米松对视网膜血管通透性以及血管内皮生长因子(vascular endothelial growth factor,VEGF)表达的影响。方法将大鼠分为对照组、糖尿病组、糖尿病+生理盐水组和糖尿病+地塞米松4组。分别用伊文思蓝方法检测血管通透性改变情况,以荧光定量PCR和酶联免疫吸附实验检测视网膜中VEGF的表达。结果糖尿病大鼠造模后4周,玻璃体内注射地塞米松(5g.L-1)。2d后与糖尿病组和糖尿病+生理盐水组相比,糖尿病+地塞米松组伊文思蓝的渗漏量分别减少了约38.2%和35.7%(P均<0.01)。与其对血管通透性的作用相一致,地塞米松显著抑制了VEGF的表达,VEGF的mRNA和蛋白表达分别减少了53.3%、52.3%、47.8%、49.5%.结论本实验提示地塞米松对血-视网膜屏障通透性的影响,至少部分是通过抑制VEGF的表达起作用。Objective To explore the effect of vasctdar endothelial growth factor (VEGF) on blood-retinal barrier breakdown in diabetic retinopathy and investigate the inhibiting effect of dexamethasone with intravitreal injection on retinal vascular permeability and VEGF. Methods AU rats are divided into four groups,including control group,diabetic group,diabetes with physiological saline group and diabetes with dexamethasone group. Retinal vascular permeability was measured by the Evans blue assay. The mRNA and protein level of VEGF were evaluated by real-time quantiative PCR analysis and enzyme-linked immunosorbent assay, respectively. Results Four weeks after streptozotocin administration, intravitreal injection of dexamethasone (5 g · L^-1) was performed. At two clays after intravitreal injection,the Evans blue leakage of diabetes with dexamethasone group were decreased 38.2% and 35.7% comparing with diabetic group and diabetes with physiological saline group(P 〈0.01 ) ,the expression of VEGF and mRNA decreasing about 53.3% , 52.3% and 47.8% , 49. 5% ,respectively. Conclusion The effect of dexamethasone on retinal vascular permeability is mediated,at least in part,through blockade of VEGF overexpression.
