摘  要:目的探讨灵杞黄斑颗粒对光损伤大鼠视网膜游离氨基酸类神经递质水平的影响。方法将42只大鼠随机分为正常对照组、阳性对照组、中药组,每组14只。建立光损伤模型,于光照后14d作组织病理学检查,并计数视网膜外核层感光细胞层数;运用柱前衍生高效液相色谱方法测定各组大鼠视网膜游离氨基酸的变化。结果光照后14d,阳性对照组和中药组视网膜外核层感光细胞层数均减少,阳性对照组明显减少,与中药组比较有显著性差异(P<0.01);阳性对照组和中药组大鼠视网膜谷氨酸(Glu)、天门冬氨酸(Asp)、甘氨酸(Gly)、牛磺酸(Tau)、γ-氨基丁酸(GABA)含量均增高,与正常对照组比较,阳性对照组增高明显(P<0.01),而中药组无明显差异(P>0.05);中药组与阳性对照组比较,视网膜5种游离氨基酸含量均较低,两者差异有统计学意义(P<0.01)。结论灵杞黄斑颗粒对光损伤引起的视网膜形态改变有保护作用,可能与抑制视网膜游离氨基酸水平的增高有关。Objective To investigate the effects of “Lingqi Huangban Granule” (LQHB) on the amino acid-like neurotransmitters in the rats with retinal light impairment. Methods Forty-two SD rats were divided into normal control group, positive control group and herbal group, whose retinal light impairment was induced by exposure to constant light for 5 hours. LQHB was given 10 days before light exposure until the animals were sacrificed in herbal group and equal volume distilled-water for the rats in positive control group. Fourteen days after light exposure, histopathological examination was carried out and the rows of outer nuclear layers were counted. The levels of glutamate(Glu), aspartate(Asp), glycine(Gly), taurine(Tau) and γ-aminobutyric acid(GABA) in the retina of rats were measured by high-performance liquid chromatography. Results Fourteen days after light exposure, the rows of outer nuclear layers were decreased, and the reduction in the positive control group was significant. There was a significant difference between the positive control group and herbal group (P〈0.01). When compared with the normal control group, the positive control group had significantly increased the levels of Glu, Asp, Gly, Tan and GABA in the retina (P〈0.01), there was no significant difference in amino acid level between the normal control group and herbal group (P〉0.05). Compared with the positive control group, the levels of amino acids had reduced significantly. Conclusion LQHB can protect the morphology changes after light exposure possibly by inhibiting the level of amino acids in the retina.
