摘　　要：目的研究PLGA(乳酸-乙醇酸共聚物)-吉西他滨缓释微球间质化疗用于胰腺癌治疗的药动学特点。方法将移植成功的胰腺癌荷瘤鼠分为2个组:PLGA-吉西他滨缓释微球瘤内给药间质化疗组和吉西他滨裸药腹腔给药全身化疗组。观察指标:半衰期(t_(1/2)),曲线下面积(AUC)、达峰浓度(tmax)。结果间质化疗与全身化疗相比较,药物代谢的半衰期显著延长,曲线下面积增加,峰浓度提高,两者相比,结果具有统计学差异,P值均小于0.01。结论间质化疗具有独特的药代动力学方面的优势,能够延长药物在局部的作用时间,提高局部的药物浓度。Objective To study the pharmacokinetics of interstitial chemotherapy with slow - release PLGA （poly lactic - co - glycolic acid） - gemcitabine microsphere. Methods Tumor - bearing nude mice were randomly divided into two groups, receiving interstitial chemotherapy and systematic chemotherapy respectively. The half time （t1/2 ） , area under the curve （AUC） and peak concentration （Tmax） were determined. Results Interstitial chemotherapy has longer half time, larger AUC, higher Tmax compared with systematic chemotherapy. There were statistical differences of these three pharmacokinetic parameters between the two groups. Conclusion Interstitial chemotherapy could prolong the regional active time and improve the regional concentration of chemotherapeutic drugs.
