细胞色素C亚细胞分布和Caspase-3活性改变在丝裂霉素C诱导膀胱癌细胞凋亡中的作用

摘  要:目的:探讨丝裂霉素C(MMC)诱导人膀胱癌细胞凋亡的分子机制。方法:不同浓度MMC作用于膀胱癌BIU-87细胞,四甲基偶氮唑蓝(MTT)比色法检测细胞存活率,吖啶橙(AO)荧光染色、流式细胞术(FCM)检测细胞凋亡,并对半胱氨酸蛋白酶3(Caspase-3)活性、细胞色素C亚细胞分布变化进行检测。结果:BIU-87细胞存活率与MMC浓度呈负相关(r=-0.97,P<0.05)。对照组和16、32、64、128μg/ml的MMC处理组凋亡率分别为(1.10±0.20)%、(10.93±1.51)%、(22.51±2.49)%、(31.71±3.39)%、(53.2±3.81)%,Caspase-3活性分别为0.152±0.009、0.201±0.010、0.275±0.009、0.479±0.012、0.594±0.013,MMC处理组与对照组相比差异均有统计学意义(P<0.05)。MMC处理组线粒体内细胞色素C含量显著低于对照组(P<0.05)。结论:促进线粒体细胞色素C释放和Caspase-3激活可能是MMC诱导膀胱癌细胞凋亡的重要机制。Objective:To study the molecular mechanism of mitomycin(MMC) to human bladder cancer cells.Methods:Different concentrations of MMC were applied to human bladder cancer BIU-87 cells,then MTT method was used to assay survival rates,fluorescent staining with acridine orange(AO) and flow cytometer(FCM) were used to detect cell apoptosis.Changes of Caspase-3 activity and intracellular distribution of cytochrome C were also detected.Results:The survival rate of BIU-87 cells expressed a negative correlation to the concentrations of C2-cer(r =-0.97,P 〈 0.05).The apoptosis rates of control group and the groups of 16、32、64、128 μg/mL of MMC were(1.10±0.20)%,(10.93±1.51)%,(22.51±2.49)%,(31.71±3.39)%,(53.2±3.81)%.The activities of Caspase-3 were 0.152±0.009,0.201±0.010,0.275±0.009,0.479±0.012,0.594±0.013,respectively.The differences between the groups of MMC and control group were statistically significant(P 〈 0.05).Compared with control group,the amount of cytochrome C in mitochondria was less in the groups of MMC(P 〈 0.05).Conclusion:MMC may induce apoptosis of human bladder cancer BIU-87 cells by promoting the release of cytochrome C from mitochondria and activating Caspase-3.
