摘  要:[目的]为工业提取麦冬总皂苷选出一种新的提取方法。[方法]以短葶山麦冬皂苷C为标准品,利用等吸收双波长消去法、薄层层析法、高效液相色谱法进行含量分析,对生物酶法和常规醇提法提取的麦冬总皂苷进行比较研究。[结果]在2-15μg/ml浓度范围内,麦冬皂苷浓度(C)与吸收差值(△A)成线性关系,其线性回归方程为:△A=-0.005 51+6.532 45C,R=0.999 21。3种方法的测定结果一致表明生物酶法提取的麦冬总皂苷浓度较常规醇提法的大。等吸收双波长消去法测得生物酶法提取的麦冬总皂苷得率为5.66%,而常规醇提法的仅为2.36%。生物酶法提取的短葶山麦冬皂苷C的浓度为0.081 34 mg/ml,是常规醇提法(0.039 06 mg/ml)的2.08倍。[结论]采用生物酶法提取麦冬总皂苷可以提高提取得率。[Objective]The study aimed to select out a new method for the industrial extraction of total saponin from Ophiopogon japonicus.[Method]With the saponin C from Liriope muscari(Desne) Baily as standard,the content analysis was conducted by equal absorption double wavelength spectrophotometry,TLC and HPLC and the total saponin extracted from O.japonicus by biological enzyme method was compared with that by conventional alcohol extraction.[Result]In the concn.range of 2~15 μg/ml,the O.japonicus saponin concn.(C) showed linear relationship with absorption difference value(△A) and their linear regression equation was △A=-0.005 51+6.532 45 C,R=0.999 21.The determination results of the 3 methods accordantly showed that the concn.of total saponin extracted from O.japonicus by biological enzyme method was bigger than that by conventional ethanol extraction.The yield of total saponin extracted from O.japonicus by biological enzyme method and determined by equal absorption double wavelength spectrophotometry was 5.66%,and that by conventional ethanol extraction was only 2.36%.The concn.of saponin C extracted from L.muscari by biological enzyme method was 0.081 34 mg/ml and 2.08 times of that by conventional ethanol extraction(0.039 06 mg/ml).[Conclusion]Extracting total saponin from O.japonicus by biological enzyme method could enhance the extracting yield.
