差异表达基因整合素α_2白细胞介素-8 Cx43在早期宫颈鳞癌中的表达及意义

摘　　要：目的研究早期宫颈鳞癌差异表达基因整合素α2、白细胞介素-8(IL-8)和间隙连接蛋白-43(Cx43)在宫颈鳞癌中的表达及其与淋巴结转移的关系,探讨其在宫颈鳞癌淋巴结转移中的作用机制。方法采用反转录-聚合酶链反应(RT-PCR)、组织芯片进行的免疫组织化学方法检测早期宫颈鳞癌组织中整合素α2、IL-8和Cx43mRNA及蛋白的表达。结果有淋巴结转移的早期宫颈鳞癌组织中整合素α2和IL-8的表达显著高于非转移组织(P<0.05),CX43的表达则相反(P<0.01)。且整合素α2和IL-8的表达呈正相关(r=0.241,P<0.05)。结论整合素α2、IL-8和CX43与早期宫颈鳞癌的淋巴结转移密切相关,在一定程度上发挥了促进和抑制早期宫颈鳞癌淋巴结转移的作用。Objective To determine the correlation between expressions of Integrin a2, Cx43 , interleukin 8 （IL-8） and lymph node metastases in earlier cervical squamous cancer and to study the mechanism of early metastasis. Methods Integrin a2, IL-8 and Cx43 mRNA expressions were examined with reverse transcription polymerase chain reaction（RT-PCR） and tissue microarray to determine the expression of their protein. Results The cervical cancers with lymph node metastases expressed a2 and IL-8 at significantly higher levels than those with negative lymph nodes （P〈0.05）.The contrary was seen in CX43 expression （P〈0.01）. Moreover, Integrin a2 and IL-8 expressions were shown to be the strongly correlated in cervical cancer （r=0.241,P〈0.05）. Conclusion There was a positive correlation between Integrin a2, IL-8 and CX43 expressions and lymph node metastases of cervical cancer. Activities of Integrin a2, IL-8 and CX43 may be somehow involved in the early spread of cervical cancer cells to lymph nodes.
