摘 要:目的探讨COPD(慢性阻塞性肺疾病)患者血清MMP-9、TIMP-1比率与气流阻塞的关系。方法应用酶联免疫(ELISA)方法检测88例COPD患者和40名正常人血清中MMP-9、TIMP-1浓度。结果COPD患者的MMP-9、TIMP-1血清浓度(ng/mL)(130.66±101.16)、(235.16±112.34)明显高于对照组(32.71±16.25)、(142.77±43.60),差异具有显著性(P<0.01);COPD组及对照组血清MMP-9、TIMP-1浓度与FEV1占预计值百分比具有明显负相关(相关系数r=-0.601、-0.621,P<0.01);COPD组及对照组血清MMP-9、TIMP-1浓度与FEV1/FVC%具有明显负相关(r=-0.562、-0.661,P<0.01);MMP-9/TIMP-1比率与FEV1占预计值%及FEV1/FVC%具有明显负相关(r=-0.396、-0.372,P<0.05)。结论COPD患者血清及MMP-9、TIMP-1浓度及MMP-9/TIMP-1比率增加,与气流阻塞成负相关。MMP-9、TIMP-1是引起气流阻塞很重要的因素。[Objective] To investigate the alteration of MMP-9, TIMP-1 expression in serum from patients with COPD, as well as the correlation between the expressions of these factors and lung function. [Methods] Enzymelinked Immunosorbent Assay (ELISA) was carried for the levels of MMP-9 and TIMP-1, in serum from 88 patients with COPD and 40 samples from health donors. [Results] The serum levels of MMP-9, TIMP-1 (130.66±101.16) ng/mL, (235.16±112.34) ng/mL from patients with COPD were statistically significantly higher than the control group (32.71±16.25) ng/mL, (142.77±43.60) ng/mL, P 〈0.01. There was a significant inverse correlation between the levels of MMP-9, TIMP-1 in serum and FEVI% (r =-0.601, -0.621), respectively, P 〈0.01). This inverse correlation was also seen between MMP-9, TIMP-1 and FEV1/FVC% (r =-0.562, -0.661, respectively, P 〈0.01). Furthermore, inverse relationships were observed between the ratio of MMP-9/TIMP-1 and FEVI%, as well as MMP-9/TIMP-1 vs. FEV1/FVC% (r =-0.396, -0.372) respectively, P 〈0.05). [Conclusions] There were significant elevations of MMP- 9 and TIMP-1 in serum of patients with COPD, and increased ratio of MMP-9/TIMP-1 inversely correlates with air- flow obstruction. These results suggested that MMP-9 and TIMP-1 might play important role in development of airway obstruction.
