摘  要:目的观察美满霉素(minocycline)对血管性痴呆大鼠脑组织和血清血管内皮生长因子(VEGF)I、L-1β、和TNF-α表达的影响,探讨美满霉素对血管性痴呆脑保护作用机制。方法Wistar大鼠随机分组:正常组(N组)、假手术组(S组)、痴呆模型组(M组)、美满霉素治疗组(MT组)。酶联免疫吸附法检测大鼠脑组织VEGFI、L-1β、TNF-α表达水平和血清水平。结果MT组IL-1β、TNF-α表达较M组降低(P<0.05),MT组VEGF表达较M组增高(P<0.05);MT组VEGFI、L-1β、TNF-α表达较N、S组增高(P<0.05);M组VEGFI、L-1β、TNF-α表达较N、S组显著增高(P<0.01)。结论美满霉素能降低血管性痴呆大鼠脑组织和血清IL-1β、TNF-α水平、增强VEGF表达,美满霉素可以通过抑制血管性痴呆大鼠神经炎性反应和增强血管修复发挥脑保护作用。Objective To investigate the minocycline influences on expression of VEGF,IL-1β and TNF-α for in vascular dementia brain and serum level of VEGF,IL-1β and TNF-α in vascular dementia rats, to study neuroprotection mechanism of minocycline for vascular dementia. Methods Wistar rats were randomly divided into: normal group (N), sham group (S), vascular dementia model group (M) and minocycline treatment group (MT). Serum level, expression of VEGF,IL-1β and TNF-α were measured by enzyme linked immunosorbent assay. Results The serum level,expression of IL-1β and TNF-αin MT groups were lower than that of in M (P〈0.05) ,the serum level and expression of VEGF in MT groups were higher than that of in M (P〈0.05); the serum level and expression of VEGF, IL-1β and TNF-α in MT and M groups were higher than that of in N,S(P〈0.01). Conclusion Minocycline can restrain the expression of IL-1β , TNF-α and increase the expression of VEGF in vascular dementia, minocycline can enhance recovery of cerebral vascular to protect neural function by restraining neuroinflammation.
