摘 要:目的:兔VX2肿瘤细胞株可接种在兔的肝、肾、骨及脑等部位制成肿瘤模型,但是接种于肌肉内建立的肿瘤模型很少用于实验研究。建立兔VX2肌肉肿瘤模型并观察其生物学特性。方法:实验于2007—03/08在重庆医科大学医学生物工程研究所完成。①实验材料:两三个月月龄纯种新西兰大白兔33只,雌雄不限,体质量1.5~2.0kg;VX2肿瘤细胞株由日本京都Funabashi Farm公司馈赠。②实验过程:采用组织块包埋法,将VX2肿瘤组织块移植于33只新西兰大白兔大腿股外侧肌内建立软组织肿瘤模型。③评估:随机取15只荷瘤兔,分别在接种肿瘤后7,10,14,21,28d行B超、彩色多普勒血流成像检查;另18只分别于接种后10,15,20,25,30,35d随机麻醉后处死3只荷瘤兔,进行大体解剖和病理学检查。实验过程中对动物处置符合动物伦理学标准。结果:33只兔均进入结果分析。①建立的兔VX2肌肉肿瘤模型成瘤率高达100%,接种7~10d,肿瘤生长迅速,生长率为55.9%:接种10~14d,肿瘤生长减缓,生长率为68.6%;14d以后,肿瘤再次迅速增长。②B超示21d后肿瘤内部可见形状不规则,大小不等的小片状无回声区(9/15,60%)或高回声钙化区(4/15,26.7%);彩色多普勒血流成像肿瘤边缘血供丰富。③病理组织学可见肿瘤细胞呈巢状或条索状弥漫分布,瘤细胞体积较大,形态不规则,有较多核分裂相,细胞异型性大。④肿瘤移植后25d开始出现肺转移,其他部位均无转移。结论:此模型移植成功率高,建模时间短且肿瘤生长迅速,肿瘤血供丰富,转移发生较晚,且只发生肺转移,无其他部位转移,该模型是一种较为理想的用于研究肿瘤局部治疗对肺转移影响的动物模犁。AIM: Rabbit VX2 tumor cell strain can be transplanted in liver, kidney, bone and brain of rabbits to establish tumor models. Tumor model by transplanting tumor cell strain in the muscle is seldom used in empirical study. This study established a model of VX2 muscle tumor in rabbit and observed its biological features. METHODS: The experiment was performed at Medical Ultrasound Engineering Institute of Chongqing Medical University from March to August 2007. ①Thirty-three purebred New Zealand white rabbits, 2-3 months old, weighing 1.5-2.0 kg, irrespective of gender were selected. The VX2 tumor tissue mass (Funabashi Farm, Japan) was transplanted in thigh vastus lateralis of rabbits using tissue mass embedding method to establish model of soft tissue tumor. ②Fifteen rabbits were randomly selected 7, 10, 14, 21, and 28 days after transplantation, respectively to undergo B ultrasound and Doppler color flow imaging examination. Three rabbits were executed after anesthesia 10, 15, 20, 25, 30, and 35 days after transplantation for gross anatomy and pathological examination. The experimental procedure was accorded with Animal Ethical Standards. RESULTS: Thirty-three rabbits were all included in the final analysis. ①The success rate of tumor transplantation was 100%. After transplantation for 7-10 days, tumor grew quickly, and tumor growth rate was 55.9%; 10-14 days, tumor growth was retarded, and the tumor growth rate was 68.6%; after 14 days, tumor grew rapidly again. ②B ultrasound showed echo free zone with irregular shape and size (9/15, 60%) or high-level echo calcified zone (4/15, 26.7%) was found in tumor. Doppler color flow imaging showed the blood supply surrounding tumor focus was abundant. ③Pathohistological observation suggested tumor cell showed nest or strand shape. Neoplastic cells were large with irregular shape, many karyokinesis, and large cell heteromorphism. ④The tumor began to metastasize to lung on the 25th day after transplantation, and no metastases to other organs
