温州汉族群体4个Y—STR复合扩增及其单倍型多态性研究

摘　　要：目的建立DYS426、DYS393、DYS390、DYS438基因座的复合扩增体系,研究四基因座单倍型频率。方法建立复合扩增体系,利用聚合酶链式反应(PCR)、聚丙烯酰胺凝胶电泳(page)及银染分型技术,对温州地区汉族155名无关个体的DYS426、DYS393、DYS390、DYS438位点进行分型,检验其单倍型多态性。结果复合扩增DYS426、DYS393、DYS390、DYS438位点分别检出4、8、5和4个等位基因,共52种单倍型,其平均基因差异度为0.9608。结论该研究Y-STR基因座复合扩增体系和所获单倍型频率数据在法医学个人识别和亲子鉴定中有较高实用价值,适用于温州汉族群体。Objective： o establish a multiplexing Y - STR system and study haplotype frequencies of 4 Y - specific loci in Wenzhou Han population. Methods： DYS426, DYS393, DYS390 and DYS438 loci were amplified simultaneously by using a multiplex amplification system and were analyzed by polyacrylamide gel electrophoresis and silver staining. Results： 155 male unrelated individuals from the Han population in Wenzhou were tested to determine the haplotypes frequencies. Results： 4, 8, 5, 4 alleles of DYS426, DYS393, DYS390 and DYS438 loci were found, respectively. 52 different heliotypes were detected. Average gene diversity of this system was 0. 9608. Conclusion- The multiplex amplified system of these 4 Y - specific loci is useful for forensic practice.
