摘 要:目的探讨促红细胞生成素(EPO)对大鼠脑缺血再灌注后血红素加氧酶1(HO-1)表达的影响。方法随机将雄性SD大鼠分为缺血再灌注组、治疗组、假手术组。治疗组按照缺血再灌注后不同时间点给予EPO腹腔注射,假手术组不手术,于各时间点断头取脑,用逆转录-聚合酶链反应技术检测大鼠皮层HO-1 mRNA的表达。结果①全脑缺血再灌注后HO-1 mRNA的表达发生动态变化,缺血后6h开始明显升高,24 h达高峰,此后缓慢下降;②EPO能够显著上调全脑缺血后皮层HO-1 mRNA的表达,缺血再灌注后即注射EPO组与缺血再灌注组各时间点HO-1和-βactin吸光度的比值差异有统计学意义(P<0.05)。结论EPO的神经保护作用可能部分通过上调HO-1的表达来实现。Objective To explore the effect of erythropoietin(EPO) treatment on the expression of heme oxygenase-1 (HO-1 )mRNA in rats with globe ischemia-reperfusion injury, Methods The 4-vessel obstructive globe ischemic rat models were used in this study. The rots were randomly divided into three groups: the ischemia-reperfusion group, the experimental group and sham-operation group. Rats in the ischemia-reperfusion group and the experimental group were subjected to EPO injection into the abdominal cavity after globe ischemia, however rots in the sham-operation group were not subjected to an operation. The expression of HO- 1 mRNA was determined by RT-PCR. Results (1) After ischemia, the expression of HO-1 mRMA was obviously changed based on time. (2) The effect of EPO treatment on the expression of HO- 1 mRNA in the cortex of this rat model was obvious. Conclusion The protective effect of EPO for ischemia-reperfusion injury is perhaps induced by up-regulating the expression of HO-1.
