摘 要:目的检测肺鳞癌组织中p14ARF mRNA及其蛋白的表达,探讨此因子在肺鳞癌发生和发展中的作用及临床应用价值。方法采用原位杂交方法检测56例肺鳞癌和13例癌旁鳞化及炎症肺组织(对照组)中p14ARF mRNA的表达,同时采用免疫组化方法检测相应标本中p14ARF蛋白的表达。结果在肺鳞癌和对照组中p14ARF mRNA的阳性表达率分别为44.6%和84.6%,两者比较差异有统计学意义(χ2=6.756,P=0.009)。p14ARF蛋白表达的阳性率分别为41.1%和76.9%,肺鳞癌组远低于对照组,差异有统计学意义(χ2=5.435,P=0.020)。p14ARF mRNA和蛋白在肺鳞癌组织中的阳性表达具有一致性(rs=0.930,P<0.05),并且都随组织分化程度的降低而降低,高分化鳞癌与低分化鳞癌之间差异有统计学意义(P<0.05)。结论p14ARF蛋白的表达异常可能与肺鳞癌的发生、发展有关,p14ARF蛋白的检测可能为临床判断组织恶性程度提供参考。Objective To investigate the expression of the p14^ARF mRNA and protein in human squamous cell carcinoma (SSC) of the lung and its clinicopathological significance. Methods Tissue specimens were collected from 56 cases of squamous cell carcinoma of the lung, specimens were also collected from 10 cases of adjacent noncancerous tissue and 3 cases of lung inflammation as the control group. All the specimens were affirmed by pathology, p14^ARF protein was detected by immunohistochemistry on paraffin-embedded sections. In situ hybridization was used to exam the expression of p14^ARF mRNA. Results The positive rate of p14^ARF mRNA in SSC and the control group was 44.6% and 84.6%, respectively(χ2= 6. 756,P=0. 009); The positive rate of p14^ARFprotein was 41.1% and 76.9%, respectively (χ2=5. 435,P=0. 020). There was a positive correlation between p14^ARF mRNA and p14^ARF protein. (rs=0. 930, P〈0.05) and the expression of p14^ARF mRNA and p14^ARF protein in SSC was significantly correlated with the tumor stage (P〈0. 05). Conclusion The downregulation of p14^ARF expression may contribute to the carcinogenesis and development of SSC of the lung and can be a biological marker for diagnosis and classification of SSC of the lung.
