摘  要:目的:研究UPR(未折叠蛋白反应,unfolded protein response)在QBc939细胞中的作用,为临床治疗胆管癌提供新思路。方法:低剂量过氧化氢(H_2O_2)、顺铂(DDP)作用于人胆管癌QBC939细胞系,MTT法测定不同处理组细胞生长曲线,westem-blot法测定不同处理组IRElα、BiP蛋白表达情况。结果:低剂量过氧化氢诱导QBc939增殖,顺铂抑制QBC939细胞增殖,IRE1α、BiP蛋白在过氧化氢组表达强阳性。结论:UPR促进细胞增殖,对细胞起保护作用,降低顺铂的抑制细胞作用。Objective: To study the role of UPR in human cholangiocarcinoma cell (QBC939) and provide a new idea to cure cholangiocarcinoma. Methods: QBC939 cells were treated by low concentration of H202 and DDP. The growth curves of QBC939 cells were detected by MTT assay. The expression of IRElα and BiP in different groups was detected by western-blot assay. Results: Low concentration of H2O2 induced the proliferation of QBC939 cells, while DDP induced the growth inhibiting effect on QBC939 cells. High expression of IRElα and BiP protein was found in the cells induced by H2O2. Conclusion: UPR enhanced cell multiplication and depressed DDP induced the growth inhibition, which provided a protection for cell.
