摘  要:目的研究脑缺血后水通道蛋白-9(AQP9)的表达变化与血脑屏障通透性之间的关系。方法采用阻断大鼠大脑中动脉制作脑缺血的动物模型,通过免疫组化方法检测AQP9的表达,并通过检测缺血脑组织中伊文思蓝(EvansBlue,EB)外渗的量,以反应血脑屏障通透性的变化。结果对照组AQP9表达和EB含量无明显改变。与对照组相比,缺血组AQP9的表达随缺血时间的延长而表达增加;且AQP9的表达在脑缺血后2d^3d表达最强;EB含量与AQP9的表达变化相似,EB含量分别在缺血后2d和3d增加更明显。AQP9的表达与EB含量的变化呈正相关(r>0.672,P<0.05)。结论脑缺血后AQP9表达增强,血脑屏障通透性增加,提示AQP9表达上调与血脑屏障通透性的改变有关,两者可能共同参与缺血性脑水肿的形成。Objective Objective To investigate the relationship between aquaporin-9 (AQP9) expression and blood brain barrier(BBB) penetrability after cerebral ischemic in rats. Methods The model of cerebral ischemia was made by occluding unilateral middle cerebral artery(MCA) of the rats with the suture method. The expression of AQP9 was assessed by immunohistochemiscal assay. The BBB permeability of ischemic brain wasdetermined by evans blue ( EB ) extravasation method. Results The expression of AQP9 and BBB penetrability had no significant changes in control group. The expression of AQP9 and BBB penetrability increased significantly at 6h in ischemic edema tissues after cerebral ischemia, and finally reached a peak 2d - 3d after cerebral ischemia. There was a positive correlation between expressions of AQP9 and BBB penetrability( r 〉 0.672 ,P 〈 0.05 ). Conclusion Both the expression of AQP9 and BBB penetrability in ischemic tissue increases after cerebral ischemia. The increasing penetrability of BBB may be induced by the up-regulated expression of AQP9. The results indicate that the AQP9 and BBB penetrability may be significantly correlated with the ischemic edema formation.
