摘 要:目的研究黄连素(BR)对异丙肾上腺素(ISO)诱导的大鼠心肌肥厚的影响及其作用机制。方法取SD大鼠24只,随机分为正常对照组、心肌肥厚组和BR处理组,观察各组大鼠心肌肥厚指数、心肌组织结构的改变和心肌组织中游离脂肪酸(FFA)含量以及一氧化氮合酶(NOS)活性的变化。结果心肌肥厚组与正常对照组相比,大鼠心肌肥厚指数升高(P<0.01),心肌细胞直径和横截面积均增大(P<0.01),心肌组织中FFA的含量增加(P<0.01),心肌组织中结构型NOS(cNOS)活性降低(P<0.01),诱导型NOS(iNOS)活性增强(P<0.01);BR处理组与心肌肥厚组相比,大鼠全心肥厚指数、左心室心肌肥厚指数、心肌细胞直径、心肌细胞横截面积降低(P<0.01),心肌组织中FFA含量降低(P<0.01),cNOS活性增加(P<0.05),iNOS活性降低(P<0.01)。结论BR对ISO诱导的心肌肥厚的发生有预防作用。Objective It is to study the effects of berberine on cardiac hypertrophy induced by isoproterenol in rats and its functional mechanism. Methods 24 SD rats were randomly divided into 3 groups: normal control group, hypertrophy model group and BR treatment group. The cardiac indexes, the cardiomyocyte diameter, the cross section area of cardiomyocyte, the content of free fatty acid (FFA) and the NOS activity were detected and observed at last. Results Compared with the measurements in the normal control group, the cardiac indexes, the cardiomyocyte diameter, the cross section area of cardiomyocyte, the content of FFA, the activity of inducible NOS were remarkably increased in hypertrophy model group (P 〈 0.01 ), and the activity of constitutive NOS was significantly decreased (P 〈 0.01 ). Further comparison between the hypertrophy model group and the BR treatment group showed that the berberine prevented the cardiac hypertrophy induced by isoproterenol and decreased the the cardiac indexes, the cardiomyocyte diameter, the cross section area of cardiomyocyte, the content of FFA and the activity of inducible NOS( P 〈 0.01 ); berberine elevated the activity of constitutive NOS (P 〈 0.05). Conclusion Berberine can prevent the cardiac hypertrophy induced by isoproterenol.
