经阿霉素诱导的人肺腺癌A549细胞中侧群细胞比例变化及意义

摘  要:目的探讨多药耐药性与肿瘤侧群细胞之间的关系。方法以小剂量阿霉素(ADM)浓度梯度递增的方法诱导人肺腺癌A549细胞系,12周后观察细胞形态变化,用MTT法进行细胞药物敏感性检测,Hoechst33342/PI染色测定侧群细胞(SP细胞)比例。结果A549/ADM细胞以长梭形为主,胞体增大,伪足增多,对阿霉素、顺铂、长春新碱、足叶乙甙、紫杉醇5种药物的耐药指数分别为9.588、6.348、1.736、11.213、4.333,SP细胞比例增加10倍。结论肺腺癌A549细胞多药耐药性与侧群细胞比例增加有关。Objective To study the relationship between muhidrug resistance and the side population(SP) cells. Methods The human lung adenocareinoma A549 cell line was exposed to adriamyein of a low and increasing concentration for 12 weeks, then morphology of the cells was observed with microscope and the relative resistance was tested with MTT assay and the ratio of the SP cell was observed with fluorescence microscope. Results The morphology of A549/ADM cell changed obviously, The resistance factor of A549/ ADM cells to doxorubincin, cisplatin, etoposide, vincristione, paclitasel was 9. 588,6. 348,1.736,11.213,4. 333 respectively and the ratio of SP cell increase obviously. Conclusion SP cells increasing in human lung adenocarcinoma was related with the muhidrug resistance.
