HIF-1α、Caspase-3、Survivin蛋白在食管鳞癌组织中的表达及相关性分析

摘　　要：目的研究缺氧诱导因子-1α(HIF-1α.)、Caspase-3、Survivin在食管癌的发生、发展、浸润、转移过程中的作用及有无相互关系,为进一步寻找抑制食管鳞癌发生、发展的途径提供理论依据。方法应用免疫组化S-P法检测49例食管鳞癌组织中HIF-1α、Caspase-3、Survivin蛋白的表达并进行相关性分析。结果HIF-1α蛋白在食管鳞癌组织中的阳性率为51.0%(25/49),显著高于正常食管黏膜0%(0/32)(P<0.05);HIF-1α蛋白的阳性表达与Caspase-3蛋白表达呈负相关关系,与Survivin蛋白表达呈正相关关系,提示HIF-1α蛋白的阳性表达下调了Caspase-3蛋白的表达,使Survivin蛋白的表达率增加,食管癌细胞凋亡受阻。结论HIF-1α蛋白在食管鳞癌组织中的表达显著增高,联合检测HIF-1α、Caspase-3与Survivin可作为判断食管癌生物学行为的客观指标,对判断食管癌的预后具有重要意义。Objective To study of hypoxia-inducible factor-1α（ HIF-1α）protein, Caspase-3 protein and Survivin protein and their correlation in carcinogensis, development, invasion and metastasis of esophageal squamous cell carcinoma,in order to find out useful ways to suppress occurrence,development of esophageal squamous cell carcinoma. Methods Immunohistochemical S-P method detected 49 cases with esophageal squamous cell carcinoma of HIF-1α, Caspase-3 ,Survivin protein expressions and correlative analysis. Results The positive rate of HIF-1α protein in esophageal squamous cell carcinoma （ 51.0% , 25/49 ） was significantly higher than that in adjacent normal mucosa （ 0% ,0/32 ） （ P 〈 0.05）. There was a negative correlation between the expressions of HIF-1α protein and Caspase-3 protein, a positive correlation between the expressions of HIF-1α protein and Survivin protein,it implied that the positive expressions of HIF-1α protein could decrease the expres- sions of Caspase-3 protein,accordingly,ascending the expressions of Survivin protein,led to the inhibition of cellular apoptosis of esophageal squamous cell carcinoma. Conclusions HIF-1α protein in esophageal squamous cell carcinoma was significantly higher, united examining HIF-1α protein ,Caspase-3 protein and Survivin protein can become a objective target to evaluate the prognosis of esophageal squamous cell carcinoma. It had a great significance to judge the prognosis of esophageal squamous cell carcinoma.
