摘  要:目的:研究大鼠视神经夹伤后视网膜节细胞(retinal gan-glion cell,RGC)的非折叠蛋白反应(unfolded protein response,UPR)。方法:成年雄性SD大鼠28只经视神经夹伤后,在4,8,12,24,72,168h,通过免疫荧光组织化学染色法观察RGC中非折叠蛋白相关因子(PERK,IRE-1,calnexin)、AMPA受体亚基GluR2的表达;采用Fluoro-JadeC法染色显示RGC凋亡状况,并与正常对照组比较。结果:大鼠视神经夹伤后,与正常对照组相比,RGC表达PERK,calnexin明显增强。从损伤后4h,阳性RGC细胞数增加,至24h时达到峰值,PERK,calnexin阳性细胞数分别为15.00±0.36和11.75±1.44个,与对照组相比具有显著性差异(P<0.05)。从损伤后8h,RGC的IRE-1及GluR2表达增强,其中GluR2在胞膜上表达明显增强;Fluoro-Jade C染色在72h时RGC出现表达,并与GluR2的表达双标。结论:大鼠视神经夹伤后,与UPR相关的PERK,IRE-1,calnexin在RGC表达明显增强,提示UPR可能参与了RGC的凋亡。AIM:To investigate unfolded protein responses(UPR)of the retinal ganglion cell(RGC)after optical nerve crash in rats.METHODS:A total of 28 adult male SD rats were used for establishment of a model of optic nerve crash.Immunohistochemical staining was performed to detect the expressions of PERK,IRE-1 and calnexin,the related molecules of unfolded protein,and the expression of GluR2,a subunit of AMPA receptor,at 4,8,12,24,72 and 168 hours after injury.Meanwhile,Fluoro-Jade C method was adopted to show the death of RGC.The rats without any injury were served as normal controls.RESULTS:Compared with that of controls,expressions of PERK and calnexin were increased significantly(P〈0.05)in RGC after injury.The number of RGC positive to PERK and calnexin staining increased at 4 hours and reached the peak at 24 hours after injury with the values of 15.00±0.36 and 11.75±1.44 respectively.Meanwhile,expressions of IRE-1 and GluR2 increased at 8 hours and the positive staining to GluR2 was obvious on the cell membrane of RGC.Fluoro-Jade C staining in RGC was noted at 72 hours after injury,with co-existence of GluR2.CONCLUSION:The expressions of PERK,calnexin and IRE-1,the related molecules of unfolded protein,were increased obviously in RGC after optic nerve crash,indicating that UPR may be involved in the death of RGC.
