摘 要:背景与目的:脑胶质瘤治疗效果一直不理想,这与脑胶质瘤中存在一部分肿瘤干细胞有关。本研究旨在探讨脑肿瘤干细胞标记物CD133及Nestin在脑星形细胞肿瘤中的表达的临床意义。方法:采用免疫组织化学SP法检测CD133、Nestin在127例脑星形细胞肿瘤组织中的表达。结果:(1)按照WHO2000年的神经系统肿瘤分类分级标准所有标本分为Ⅰ级15例,Ⅱ级42例,Ⅲ级42例,Ⅳ级28例。(2)脑肿瘤干细胞标记物在低级别星形细胞肿瘤中低表达,在高级别星形细胞肿瘤中高表达,在低级别和高级别之间表达有显著性差异(P=0.000)。(3)CD133表达与Nestin表达之间有相关性,二者呈正相关(r=0.411,P=0.000)。(4)CD133阳细胞性率与患者性别、年龄无关(P=0.136、P=0.412),Nestin的阳性细胞率与患者性别、年龄无关(P=0.412、P=0.153)。结论:CD133及Nestin在低级别星形细胞瘤中低表达,在高级别星形细胞肿瘤中高表达。BACKGROND & OBJECTIVE: The prognosis of malignant glioma treatment is still dismal. Recent data suggest that an enriched group of stem-like cells in glioma are responsible for gliomagenesis and progression. In this paper, we explored the expression of tumor stem cell related marker in human glioma tissues. METHODS: The expression of CD133 and Nestin was examined with immunohistochemistry in 127 human astrocytoma specimens confirmed by pathology. The number and percentage of tumor cells positive for CD133 and Nestin were counted and calculated. RESULTS: (1) Among 127 tumors, 15 cases were Grade 1, 42 were Grade Ⅱ, 42 were Grade Ⅲ and 28 were Grade IV, according to WHO 2000 classification of nervous system tumors.(2)CD133 and Nestin positive cells were observed in all astrocytoma tissues. The expression of CD133 and Nestin was higher in high grade astrocytomas than that in low grade tumors(P=0.000). (3) Positive correlation was observed between CD133 and Nestin expression (r=0.411 ,P=0.000). (4) No correlation between CD133 group and Nestin expression, and sex and age, was found(P〉0.05). CONCLUSIONS: CD133 and Nestin are expressed in human astrocytomas and correlate with pathological grade.
