摘 要:目的观察硫化氢(hydrogen sulfide,H2S)对血小板L-精氨酸/一氧化氮合酶/一氧化氮(L-Arg/NOS/NO)通路的影响以探讨H2S和NO这两种气体信号分子间的相互作用。方法离体孵育大鼠血小板,加入H2S气体溶液(10-6~10-3mol/L)孵育2h,采用Greiss法测定血小板孵育液亚硝酸盐(NO2-)含量;同位素示踪法检测血小板NOS活性及L-Arg转运。结果一次性给予H2S(10-6~10-3mol/L)孵育2h,呈浓度依赖的抑制了L-Arg/NO通路,血小板孵育液中NO2-含量比对照组分别降低6%、11%(均P>0.05)、24%、36%和52%(均P<0.01);NOS活性分别下降1%、14%(P>0.05)、25%、37%和42%(P<0.01);L-Arg转运分别减少12%(P>0.05)到72%(P<0.01);IC50分别为53.02、41.63及21.53μmol/L(P<0.01)。结论H2S通过抑制血小板L-Arg转运和NOS活性,下调L-Arg/NOS/NO通路,从而减少血小板NO生成。[Objective] To observe the influence of hydrogen sulfide (H S) on L-arginine/nitric oxide synthase (NOS)/nitric oxide (NO) pathway in rat platelets, and explore the interaction between H2S and NO as cardiovascular regulatory gasotransmitters. [Methods] Washed platelets ( 1 ×10^8 pt/mL) was incubated with H2S (10^-6 mol/L-10^-3 mol/L) for 2 h, the nitrite production was measured by Greiss assay, NOS activities and L-arginine transport rate were determined by isotope tracer method. [Results] After the incubation for 2 h, H2S (10^-6 mol/L-10^-3 tool/L) inhibited the L-arginine/NOS/NO pathway in rat platelets in a dose-dependent manner, the nitrite production of platelets decreased by 6%, 11%(P〉0.05), 24%, 36% and 52%(P〈0.01), NOS activity reduced by 1%,14%(P〉0.05), 25%, 37% and 42%(P〈0.01), L-arginine transportation decreased by 12%(P〉0.05)to 72%(P〈0.01) and their IC50 were 53.02 μmol/L, 41.63μmol/L and 21.53μmol/L(P 〈0.01), respectively. [Conclusion] H2S inhibits NO release by inhibiting L-arginine transport and NOS activity, down-regulating L-arginine/NOS/NO pathway in rat platelets.
