摘 要:[目的]探讨Trichoderma reesei外切葡萄糖苷酶CBHⅠ对木糖是否有糖苷合成活性。[方法]以木糖为底物,检测了155min反应体系中还原糖含量的变化,并以GC/MS对反应产物进行定性分析。结果]在封闭的反应体系中,还原糖含量出现规律的周期性上升下[降的特征,在2条回归曲线y=3E-05x+0.7151和y=-0.0002x+0.6276之间呈波状振荡,还原糖含量振荡周期为(33.75±4.79)min,从而推断出CBHⅠ可以以木糖为底物合成非还原性产物。对非还原性产物衍生后进行GC/MS分析,产物中有以木糖为单位的非还原性二糖存在。[结论]CBHⅠ对木糖有糖苷合成活性,这扩大了其在糖苷合成中底物的应用范围。[Objective] The aim was to explore whether the CBH [ from Trichoderma reesei had the activity of glycoside synthesis with xylose as the substrate. [Method] The content change of reducing sugar in the reaction system was detected in 155 min and the reaction products was analyzed by GC/MS. [Result] In the close reaction system, the reducing sugar content changed regularly with periodic raise and decrease. The reducing sugar content showed wavy oscillation between y = 3E-05x + 0.7151 and y = -0.0002x + 0.6276, and its oscillation period was ( 33.75±4.79)min. So it was speculated that CBH Ⅰ could catalyze the synthesis of non-reducing matter with xylose as substrate. By means of GC/MS, it was confirmed that the non-roducing matter was a kind of disaccharide. [Conclusion] CBH Ⅰ from Trichoderma reesei had the glycoside synthesis activity with xylose as the substrate, which enlarged the application range of substrate in glycoside synthesis.
