摘 要:目的比较人乳腺癌原代细胞以及人乳腺癌细胞系MCF-7、MDA-MB-231S中乳腺癌干细胞的含量和细胞表型。方法采用流式细胞技术对人乳腺癌原代细胞以及MCF-7、MDA-MB-231S进行检测和分选,并测定不同细胞亚群在裸鼠体内重建肿瘤的能力。结果人乳腺癌原代细胞以及MCF-7、MDA-MB-231S细胞的CD44和CD24表达与分布不同;和人乳腺癌原代细胞相比,MCF-7、MDA-MB-231S细胞中乳腺癌干细胞的含量更高(P<0.01)。结论和人乳腺癌原代细胞相比,人乳腺癌细胞系中含有更高比例的乳腺癌干细胞,并且细胞表型也发生了改变。Objective To compare the content and phenotype of cancer stem cells in the MCF-7, MDA-MB-231S human breast cancer cell line with those in the human breast cancer primary cells. Methods Different subpopulation cells were detected and separated from MCF-7, MDA-MB-231S and human breast cancer primary cells by flow cytometry,and their ability to reconstruct tumor was determined in vivo. Results The expression and distribution of CD44 and CD24 in MCF-7,MDA-MB-231S and human breast cancer primary ceils were different. Compared with human breast cancer primary cells,the proportion of cancer stem ceils in MCF-7 and MDA-MB-231S ceils was higher (P 〈 0.01 ). Conclusion Compared with human breast cancer primary ceils, there am more breast cancer stem cells in the human breast cancer cell line and the phenotype has been changed.
