摘  要:目的探讨RASSF1抑癌基因两种不同转录子RASSF1A、RASSF1B在脑胶质瘤中的表达情况及RASSF1与临床病理因素之间的关系。方法采用逆转录聚合酶链式反应(RT-PCR)检测46例脑胶质瘤组织及6例脑正常组织中RASSF1A、RASSF1B mRNA的表达情况。结果RASSF1A在6例正常脑组织标本中完全表达。在脑胶质瘤组织中的表达缺失率为47.8%(22/46),两组之间比较差异有显著性(P<0.01)。RASSF1B在正常脑组织中可完全表达,在脑胶质瘤组织中的表达缺失率为26.1%(12/46)。两组之间比较差异无显著性(P>0.05)。RASSF1A、RASSF1B在胶质瘤中表达缺失率与各病理组织学类型及分化程度之间比较差异无显著性。结论RASSF1A、RASSF1B在脑胶质瘤中的表达缺失程度不同,RASSF1A表达缺失更明显。RASSF1A、RASSF1B在胶质瘤中表达缺失率与各病理组织学类型及分化程度无关。检测RASSF1A在脑胶质瘤中的缺失情况对临床判断肿瘤发生有一定的指导意义。Objective To investigate the expression of two different RASSF1 transcripts and its clinical relations with the glioma pathology. Methods The mRNA expression of RASSF1A and RASSF1B was detected by RT-PCR method in 46 specimens of brain glioma and 6 normal brain tissues. Results The expression of RASSF1A and RASSF1B was complete in six normal brain tissuesoThe rates of expression loss of transcripts A and B in glioma were 47.8 %(22/46)and 26.1%( 12/46)o The mRNA expression level of RASSF1A was significantly lower in gliomas than in normal brain tissues. The rates of expression loss of RASSF1A and RASSF1B mRNA had no signifcant relation with differentiation stage and pathological histology of gliomas. Conclusion The,'e is a significant difference in the expression between the two different RASSF1 transcripts of A and B in the brain gliomas, and normal brain tissues. It is important to detect the rate of expression loss of RASSF1A in the glioma.
