合贝爽减轻缺氧所致血管内皮细胞线粒体膜电位变化

摘  要:目的观察缺氧对人大血管内皮细胞线粒体膜电位的影响以及合贝爽的保护作用。方法将培养的血管内皮细胞分为3组:对照组,单纯缺氧组和缺氧加合贝爽组。应用激光扫描共聚焦显微镜定量测量各组血管内皮细胞线粒体膜电位值。结果3组血管内皮细胞线粒体膜电位(荧光强度)分别为36.578±1.147,28.142±1.124,33.756±1.124。①与对照组相比,单纯缺氧组的线粒体膜电位水平显著降低(P<0.01)。②与单纯缺氧组相比,合贝爽加缺氧组内皮细胞线粒体膜电位水平有显著升高(P<0.01),接近对照组测定值(P>0.05)。结论缺氧引起血管内皮细胞线粒体膜电位降低;合贝爽可减轻缺氧损伤所致的线粒体膜电位的降低,从而具有稳定线粒体膜电位的作用。Objective To study the effect of hypoxia on intracellular mitochondria membrane potential (MMP)of human vascular endothelial cells and the protective effect of diltiazem. Methods Cultured vascular endothelial cells were divided into 3 groups: control group, hypoxia group,and combination group(combined dihiazem with hypoxia). The changes of intracellular MMP were detected using laser scanning confocal microscope. Results ①Compared with control group, the intracellular MMP in hypoxia group significantly decreased(P〈 0.01 ). ②Compared with hypoxia group, the intracellular MMP in combination group significantly increased (P〈0.01), but close to that in control group (P 〉 0.05). Conclusion Hypoxia could significantly influence the intracellular MMP, but diltiazem can attenuate the hypoxia-induced decrease of intracellular MMP.
