PDGF-AA和PDGF-BB在大鼠矽肺组织中的表达及意义

摘　　要：目的研究血小板衍生生长因子-AA(PDGF-AA)和血小板衍生生长因子-BB(PDGF-BB)在二氧化硅所致矽肺大鼠肺组织中的表达。方法40只SD大鼠随机分为模型组和对照组,每组20只,雌雄各半。模型组大鼠向气管灌注石英粉尘制作矽肺模型;正常对照组以灭菌生理盐水lml代替。45d后,观察各组大鼠肺组织的病理改变,免疫组化方法观察肺组织中PDGF-AA和PDGF-BB的蛋白表达情况。结果PDGF-AA和PDGF-BB在实验组矽肺模型肺组织中的表达量均高于在正常对照组大鼠肺组织中的表达量,且差异有统计学意义(P<0.05);两者蛋白表达量间无直线相关关系(P>0.05)。结论PDGF-AA和PDGF-BB可能参与了尘肺的发生发展。Objective To study the expression of platelet derived growth factor - AA and platelet derived growth factor - BB in lung of rats with experimenatial pneumoconiosis. Methods Total 40 SD rats were divided randomly into the experimenatial and control group, twenty rats in each group, in which female and male each occupied 50%. Silicosis rat models were established by intratracheal instillation silicon dioxide, the degerming physiological saline was used in the control group. Forty - five days later, the pathological changes were observed, the expression of platelet derived growth factor -AA and platelet derived growth factor- BB in the lung tissue was verified by immunohischemical analysis. Results The expression of PDGF - B Blevel were higher in the model group compared with that of the control group （P 〈 0. 05 ） . The correlation between protein of PDGF - AA and PDGF - BB in the two groups was negative （P 〉 0. 05）. Conclusion PDGF - AA and PDGF - BB may participate in the development of pneumoconiosis.
