摘  要:目的观察连续多次使用氯胺酮(ketamine,Ket)对小鼠体重、自主活动和学习记忆的影响。方法按分层随机区组设计将小鼠分为4组(n=10):生理盐水组(NS组,ip),氯胺酮5 mg·kg^-1、10 mg·kg^-1、20 mg·kg^-1(ip)(K5、K10、K20组)。d1-10每隔24 h给药一次,d11停药并测量停药前后小鼠体重、自主活动次数、避暗潜伏期及错误次数。d12侧脑室注射NMDA进行促发实验,并分别于NMDA注射前后测量观察指标。d13-17每隔24 h给药一次,d18、d19分别重复d11测量观察指标。结果与NS组相比,K5、K10、K20组在以上各同一时间点体重、自主活动、避暗潜伏期及错误次数的差别均无统计学意义(P〉0.05)。结论连续多次使用氯胺酮后自然戒断及用NMDA促发对小鼠体重、自主活动、学习记忆无明显影响,提示氯胺酮对小鼠无明显躯体依赖性。Objective To observe the effects of repeated ketamine (Ket) on body weight, autonomic activities, learning and memory in mice. Methods Fourty Kuming male mice were randomly divided into 4 groups of 10 animals each and treated with normal saline 0. 1 mg·kg^-1 ip, Ket 5mg·kg^-1ip, Ket 10mg·kg^-1ip and Ket 20 mg·kg^-1ip for NS, K5, K10 and K20 groups respectively. The drug was given once a day from dl to d10.10 days after the last drug injection, the body weight, frequency of autonomic activity, length of latent period and number of errors made were noticed. NMDA was administrated by intracerebroventricular (icv) injection on d12 with the indexes tested before and after the injection. Ket ip was re - administrated once a day from d13 to d17, with the indexes re - examined on d18 and d19. Results As compared with group NS, repeated Ket administration did not cause significant differences in weight, autonomic activity, length of latent period or number of errors made after natural withdrawal, nor did repeated Ket after NMDA icv (P 〉 0. 05). Conclusion These data may demonstrate that repeated ketamine does not cause significant physical dependence on the drug in mice.
