摘  要:目的:研究Wnt/Ca2+通路中Wnt5A,Wnt11基因在不同发展阶段的黑素瘤细胞中的表达水平.方法:运用RT-PCR法,检测正常黑素细胞和6种黑素瘤细胞中Wnt5A和Wnt11基因表达水平.结果:在正常黑素细胞和不同的黑素瘤细胞中,Wnt5A和Wnt11基因的表达均表现出动态变化的特征.Wnt5A和Wnt11水平在正常黑素细胞中表达很低,而在黑素瘤细胞中有不同程度的升高.另外,分别来自表面生长阶段、结节状生长阶段和黑色素瘤淋巴结转移的WM793B,451Lu和A2058细胞系中,Wnt5A和Wnt11基因表达水平逐渐下降,甚至在A2058细胞系中,两种基因的表达均低于正常黑素细胞.结论:在黑素瘤的不同发展阶段,Wnt5A和Wnt11基因表达的动态变化使非经典的Wnt/Ca2+通路激活程度不同.AIM: To investigate the expression levels of WntSA and Wntl I genes in Wnt/Ca^2+ signal pathway activated in different stages of melanoma progression. METHODS : By using RTPCR, we detected the expression levels of WntSA and Wntl1 genes in cultured normal epidermal melanocytes and 6 skin melanoma cell lines. RESULTS: RT-PCR results showed that the characteristics of WntSA and Wntl 1 expression were similar, both of which were dynamically changed in different cells. Compared with the low levels in normal melanocytes, WntSA and Wntl1 were up-regulated with different intensities in the most investigated melanoma cell lines except for A2058. Interestingly, WM793B, 451Lu and A2058 cell lines, which respectively derived from primary superficial spreading situation, nodular melanoma, and lymph node metastases, gradually decreased the WntSA and Wntl1 expression levels. CONCLUSION: During different stages of melanoma progression, dynamic change of WntSA and Wntl 1 gene expressions activates noncanonical Wnt/ Ca^2+ pathway with different degrees.
