摘 要:目的评价骨髓间充质干细胞(bone marrow-derived mesenchymal stem cells,MSCs)对胰岛血管重建和改善胰岛功能的作用。方法随机将SD大鼠分成2组:联合移植组,胰岛和MSCs联合移植入受体糖尿病大鼠的肾包膜下;对照组,仅作单纯胰岛移植。采用细胞密度梯度分离(Percoll)法从同种供体SD大鼠骨髓中分离得到MSCs,并进行原代培养。进一步将MSCs和分离纯化的胰岛联合植入受体糖尿病大鼠的肾包膜下。胰岛的微血管密度(MVD)通过Lectin From Bandeiraea Simplicifolia(BS-1)的荧光染色来评估,胰岛功能通过测定血糖水平来评估。结果分离培养的原代MSCs在培养的第10天后细胞开始加快增殖,细胞排列成漩涡状。于术后14天,取出胰岛移植物,作荧光BS-1染色。荧光BS-1定位于血管内皮上。联合移植组的胰岛内MVD平均为20±1.9,明显大于对照组的14±1.8(p<0.05)。联合移植组受体糖尿病大鼠的血糖缓解时间为10.5±4.1天,对照组为8.5±2.5天,两组之间无显著差异(p>0.05)。结论大鼠MSCs在胰岛移植早期有助于增加移植胰岛内的血管密度,可作为改善胰岛移植效果的新策略。Objective To To evaluate the effect of bone marrow masenchymal stem cells (MSCs) on vascular reconstruction and function improvement of transplanted pancreatic islets in rats. Methods Eight SD rats were randomly divided into two groups: pancreatic islets and MSCs transplanted group in which both of the two were implanted into kidney subcapsule of diabetic rats, normal control group in which only the pancreatic islets were implanted. MSCs were isolated from bone marrow of SD rats by Percoll density gradient centrifugatien technique; then the primary culture in vitro was done. MSCs were implanted into the kidney subcapsule of diabetic rats with pancreatic islets via isolation and purification. Microvascular density (MVD) of transplanted pancreatic islets was evaluated by staining of leetin from Bandeiraea simplicifolia (BS-1) and the function was evaluated by blood glucose level, respectively. Results The proliferation of primarily cultured MSCs was lastex horn the tenth day of culture, some MSCs arranged in whirlpool. The MVD was 20 ±1.9 in pancreatic islets and MSCs transplanted group, which was obviously higher than in normal control group which the MVD was 14 ±1.8 in (P 〈0.05). The recovery time of bloodglucese was 10.5 ± 4.1 in pancreatic islets and MSCs transplanted group and 8.5 ±2.5 in normal eentrol group, respectively. There was no significant difference between the two groups (P〉 0.05). Conclusions Combined transplantation of MSCs and pancreatic islets could be a good strategy in improvement of transplantation effect for its help in increasing the vascular density of transplanted pancreatic islets in early time.
