摘  要:目的建立体外培养细胞胶原合成模型并研究三七总苷对胶原合成的影响。方法用不同浓度和比例的新生牛血清和大鼠血清混合,筛选刺激HSC-T6细胞胶原合成的最佳条件,继而用3H-Proline掺入法研究三七总苷对其胶原合成的影响。结果10%新生牛血清加3%大鼠血清为刺激HSC-T6细胞胶原合成最佳培养条件。培养48和72h后,0.01,0.05,0.25,1.00和1.25mg·mL-1浓度的三七总苷对HSC-T6细胞胶原合成有明显的抑制作用(P<0.05);并呈剂量效应关系和时间效应关系,方差分析,F分别为4.517和5.832(均P<0.05)。结论成功建立了体外培养细胞胶原合成模型,三七总苷对细胞胶原合成有明显抑制作用。Objective To establish a model of collagen synthesis in vitro and to probe the effect of PNS on that. Methods HSC-T6 cells were incubated with the mixed sera containing different concentrations of NBS and rat sera, so as to establish a mixed sera-driven collagen synthesis model. PNS was studied on the collagen synthesis model by 3^H-Proline incorporation method. Results 10% NBS with 3% rat serum was the best condition for stimulating the collagen synthesis in HSC-T6 cells . After 48 and 72 hours'incubation, 0. 01,0. 05 ,0. 25 ,1. 00 and 1.25 mg·mL^-1 PNS significantly suppressed the collagen synthesis in HSC-T6 cells (P 〈 0.05 ) in a concentration and time dependent manner( F = 4. 517 and 5. 832, P 〈 0.05 ). Conclusion A mixed sera-driven collagen synthesis model has been successfully established, and PNS markedly inhibits collagen synthesis in HSC-T6 cells.
