摘 要:目的:探讨富含亮氨酸和精氨酸的不平衡氨基酸肠内营养(EN)联合5-氟尿嘧啶(5-Fu)对大鼠Walker-256癌肉瘤生长的影响。方法:SD大鼠空肠造瘘,皮下接种Walker-256癌肉瘤,36只接种成功的大鼠随机分为A组(平衡氨基酸+生理盐水组)、B组(平衡氨基酸+5-Fu组)、C组(富含亮氨酸及精氨酸的不平衡氨基酸+5-Fu组)。测定各组治疗后肿瘤重量、抑瘤率、PCNA指数、凋亡指数、肿瘤细胞周期及大鼠生存时间。结果:A、B、C组G0/G1期比例依次增高(P<0.01);凋亡指数依次增高(P<0.05或P<0.01);生存时间依次延长(P<0.05或P<0.01);S期比例依次下降(P<0.01);PCNA指数依次下降(P<0.01);瘤重依次下降(P<0.01)。结论:富含亮氨酸及精氨酸的不平衡氨基酸EN能明显抑制肿瘤生长,并能增强5-Fu的抗肿瘤效应。Objective To investigate the effects of imbalanced amino acid rich in arginine and leucine (IAA) enteral nutrition plus 5-fluorouracil (5-Fu) on proliferation of Walker-256 sarcocarcinoma cell line in rats. Methods The SD rats were given catheterization of jejunostomy and inoculation of Walker-256 sarcocarcinoma cells subcutaneously. Thirty-six rats successfully inoculated were randomized into 3 groups, Group A (balance amino acid plus NS), Group B (balance amino acid plus 5-Fu), Group C (IAA plus 5-Fu). Tumor weight, tumor inhibitory rate, PCNA index, apoptosis index, tumor cell cycle, and survival time were determined. Results The G0/G1 phase cell ratio, apoptosis index and survival time increased and the S phase cell ratio, PCNA index and tumor weight decreased by turns from Group A to Group C (P 〈 0.05 or P 〈 0.01). Conclusion IAA enteral nutrition can inhibit tumor growth and enhance the anti-tumor effect of 5-Fu.
