摘 要:目的探讨在氧化应激反应条件下,视网膜色素上皮(RPE)细胞对血小板源性的生长因子(PDGF)诱导的细胞迁移的影响。方法对原代培养的RPE细胞经氧化应激反应诱导剂(TBH)处理后,采用改良的Boyden腔测定在PDGF存在的情况下细胞的迁移。RPE细胞的存活用MTT比色法测定,而RPE细胞的死亡以胎盘蓝染色测定。结果PDGF能显著增加RPE细胞的迁移,RPE细胞经TBH处理后,尤其是在高浓度TBH存在的情况下,RPE细胞迁移明显受到抑制。短时间的TBH处理对RPE细胞的存活没有影响。结论TBH诱导的氧化应激反应能抑制培养条件下RPE细胞的迁移。Objective To determine if platelet derived growth factor (PDGF)-induced Retinal pigment epithelium (RPE) migration could be modulated by oxidant stress. Methods Cell migration was tested using passage 2-4 human RPE cells in a modified Boyden Chamber Assay. Cells were treated with different concentrations of tert-butyl hydroperoxide(TBH) (1-200uM) for one hour, and chemotaxis toward PDGF (30ng/ml) was measured after additional 5 hours incubation. RPE cell survival were determined using the MTT assay and RPE cell death were determined using the Trypan blue assay after exposed to tbh. Results RPE cell migration induced by PDGF was significantly increased, and induced by PDGF was significantly inhibited by pretreatment with tbh at a dose of 50uM and above. MTT assay and Trypan blue staining showed no evidence of cell death during the 6 hours of this assay. Conclusions Oxidant stress produced with tbh can inhibit PDGFinduced RPE cell migration.
