摘 要:目的观察Treg细胞对细胞毒性T淋巴细胞(CTL)抗胃癌效应中释放穿孔素、干扰素的影响。方法构建CTL细胞对MFC胃癌细胞有效杀瘤体系,加入0.5×10^5到50.0×10^5不同剂量的Treg细胞,观察CTL杀瘤活性抑制并检测穿孔素、干扰素浓度。结果Treg数量达到5.0×10^5后CTL杀瘤活性受明显抑制,穿孔素、干扰素浓度明显降低(P〈0.05)。结论Treg可通过降低CTL细胞分泌穿孔素、干扰素而抑制其杀瘤活性。Objective To investigate the in vitro inhibitory effect of Treg on restraining the CTL's anti-gastric cancer perforin and IFN-γ releasing. Methods MFC tumor killing system was established in vitro with CTL. Treg in different dosages (0.5 × 10^5 to 50.0 × 10^5 ) was added. The inhibitory effect was studied by monitoring the changes of perforin and IFN-γ. Results The killing effect was obviously suppressed when Treg reached the amount of 5.0 × 10^5. Levels of perforin and IFN-γ were significantly decreased (P 〈 0.05). Conclusion Treg can suppress antitumor effect of CTL by restraining perforin and IFN-γ releasing.
