摘 要:目的探讨益气活血复方芪丹通脉片(QDTMT)对缺血再灌注大鼠心电图及心肌中核因子-κB(NF-κB)表达的影响。方法将48只SD大鼠随机分为假手术组、模型组、芪丹通脉片低剂量组(0.36g/kg)、芪丹通脉片中剂量组(1.08g/kg)、芪丹通脉片高剂量组(3.24g/kg)及恬尔心组(5mg/kg),各组均用生理盐水配制的等体积药液灌胃,每日1次,共7d。采用冠脉结扎的方法,造成大鼠心肌缺血再灌注模型,记录心电图并采用免疫组化染色法,检测各组大鼠心肌中NF-κB蛋白的表达。结果QDTMT各剂量组和恬尔心组均能减慢心肌缺血再灌注大鼠的心率,减轻ST段抬高程度。与假手术组比较,模型组NF-κB的表达显著增多(P<0.01);与模型组比较,QDTMT各剂量组和恬尔心组NF-κB的表达显著减少(P<0.01)。结论QDTMT能抑制缺血再灌注大鼠心肌中NF-κB蛋白的表达,从而对心肌具有保护作用。Objective: To investigate the effects of Qidantongmai Tablet (QDTMT) on ECG and NF - KB in the rat with ischemia reperfusion - mediated myocardial injury (RMMI). Methods: Forty - eight SD rats were randomly divided into six experimental groups: sham operation group, control group, low dose QDTMT (0. 36g/kg) group, medium dose QDTMT (1.08g/kg) group, high dose QDTMT (3.24g/kg) group, and dihiazem hydrochloride (Smg/kg) group. Different drug or normal saline was fed to the animals by gavage at the same dosage once a day for seven days before operation. The model was established by ligating the coronary artery. Record the ECG. Tissues in the reperfused region were sampled immediately and the protein levels of NF- KB were determined by immunohistochemical staining. Results: The heart rate was stepped down and the drift of ST fragment of ECG was significantly lower QDTMT dose groups and dihiazem hydrochloride group than those in control group. Compared with that of sham operation group, the expression of NF - KB in the control group was obvious and the level of total NF - KB increased. Compared with that of control group, the expression of NF - KB was not obvious and the level of total NF- KB decreased in different QDTMT dose groups as well as the dihiazem hydrochlo ride group. Conclusion: QDTMT has cardioprotective effect against RMMI in rats possibly by inhibiting the expression of NF - KB.
