摘  要:大豆凝集素是大豆中一种主要的抗营养因子,对动物生长有一定的抑制作用,其含量作为评定动物饲料品质的重要指标。本文对血凝法测定大豆凝集素含量的方法进行了改进,并利用该方法对64份种质资源进行了凝集素含量的分析。结果表明:整粒种子粉碎过40目筛、样品与60~90℃石油醚比(W∶V)1∶10脱脂10h、样品与生理盐水(W∶V)1∶10抽提10h为宜,反应pH值在5.0~9.0为宜,反应温度在25℃左右为血凝法的最适条件,该方法具有快速鉴定、经济实用、适合大规模资源筛选等特点。64份种质资源中凝集素含量最低为1.88mg·g-1,最高为37.5mg·g-1,平均为11.91mg·g-1,85%的品种(系)在1.88mg·g-1到12.06mg·g-1之间。Soybean agglutinin ( SBA), a major anti- nutritional factor, which inhibited the growth of animals, is assessed as an important indicator of animal feed quality. Effective method for determining soybean SBA content will faeiliate the screening of soybean germplasms in large scale. In this study, Hemagglutination method of SBA was improved,with which 64 soybean germplasm from soybean Institute of Northeast Agricultural University was analyzed. Results showed the improved method for determining soybean SBA was screening crushed soybean seed with 40 mesh screens,then extracting sample for 10 h with petroleum ether( W: V) 1:10 at 60-90℃ ,and then reacting at pH 5.0-9.0 and at 25℃. In the 64 soybean accessions, the lowest,the highest, and the average content of leetin was respectively 1.88 mg·g^-1 ,37.5 mg·g^-1 and 11.91 mg·g^-1. The improved method has many advantages such as economical ,practical,and suitable for screening soybean germplasm in largescale.
